You can use both "Tumhen" (तुम्हें) and "Tum Se" (तुमसे).

I think I have heard प्यार करती हूँ used with तुम्से, not तुम्हे. Is there a difference?

Please explain the gender of love. Why in one sentence it is मेरा प्यार, and in another मेरे प्यार?

Both 'sundar' and 'khoobsooraT' are adjectives meaning 'beautiful'. You could use either of the two.
